Does anyone out there still believe that Pfu Turbo (or Ultra™) is worth it in terms of site-directed mutagenesis? Ricardipus or Hal, any ideas?
Yes, it has been a while since I've done it, but I am certain I used vanilla Pfu last time. Which, for us at least, has certain cost benefits. And I had a go at Hal a couple of months ago on this very subject, it seems.

Darned if I know. Didn't we all used to get along fine with 'vanilla' proofreading polymerases?

As a lowly honours student in a molecular bio lab, I have to say I found Pfu Turbo (and indeed the whole QuickChange kit) an absolute lifesaver.
True, that might have been as much due to the simplicity of the shake-n-bake style instructions (always a blessing to a bewildered student) as the quality of the polymerase. But still, I have a thesis because of it. And therefore worth it. Other would-be honours students take heed!

Seriously, you can do the QuikChange method without ever buying the kit, just as quickly and for less money. It's only PCR and a smidge of Dpn I, after all.
