Why does the pixelometry qualify as a result whereas the increased intensity we perceive over the DAPI staining not? Top channel blue, bottom green?

That's about the long and the short of it, yes.
The pixelometry is — or will be, when I've done a few more experiments — a quantitative measure. It eliminates one of the criticisms that can be levelled at the experiment and gives us some numbers to play with (it's actually incredibly clear in the single colour images, but there are no numbers associated with that).
The construct doesn't actually have an NLS, so it's only staying in the nucleus because it's binding to chromatin. The binding, however, is weak, so we have to do careful controls. And maybe Z series and some FRAP, just to be sure.
But yeah, we're pretty chuffed.
